IL-7 treatment consistently protects tumor-exposed T cells from CD27/CD28 loss

  • Both CTLA-4 and PD-1 regulate CD28 pathway but functionally different
    - CTLA-4 regulates the ability of CD28 to bind its ligands, whereas PD-1 regulates CD28 signaling
  • Tumor-exposed T cells undergo a 40% to 60% greater decrease in CD27 and CD28 expression than control
  • IL-7 treatment consistently protects tumor-exposed T cells from CD27/CD28 loss compared with IL-2 or IL-12
  • CD27/CD28 loss was greater in CD45RA+ (Tnavie) than in CD45RO+ (Tmem)

  • Tumor-exposed T-cells are hypoproliferative when stimulated with anti-CD3
  • IL-7 promoted proliferation in αCD-3 activated and tumor-exposed T cells
